How to read this note

1. Workflow structure
This workflow separates sample pretreatment from the shared downstream purification steps. It is intended as a practical companion to the product manual rather than a replacement for the official protocol. The downstream steps follow a magnetic bead-based purification workflow. This product also provides an automated workflow version adapted for compatible extraction instruments. For detailed setup and operation, refer to the official protocol.

2. Time interpretation
Protocol times stated in the product manual are retained where applicable. Steps without explicit timing are estimated for an experienced operator. This workflow separates active handling time from prolonged digestion time, and digestion at 55°C is the main time-determining step.

3. Workflow characteristics
This system is designed for forensic samples such as bone, teeth, hair, fingernail, seminal stains and other difficult materials. Bone and tooth samples use Buffer BGL, while other forensic materials use Buffer ATL, followed by a shared magnetic bead purification workflow.

4. Practical considerations
For small, well-prepared samples, about 3 h digestion may already provide usable DNA. For more difficult, degraded or mineralized samples, digestion can be extended up to 24 h to improve recovery. Fine bone meal preparation remains especially important for bone and teeth samples.
